The myddosome is an oligomeric signaling complex consisting of the adaptor protein MyD88 and IRAK4 kinase with stoichiometries of 7:4 and 8:4 (Watters et al, 2007; Motshwene et al, 2009).
MyD88 is a protein required for signaling of toll-like receptors with the exception of TLR3 (O'Neill and Bowie, 2007). MyD88 is required also for intracellular signaling in response to IL1 and IL18 (O'Neill, 2008). IRAK kinases (IRAK1, IRAK2, and IRAK4) are kinases activated by Toll-like receptors via MyD88.
