This hybrid protein is a chimeric tumor-targeted killer protein designed by Xu et al (2004). Immunocasp-6 comprises a single-chain antibody directed against erbB2 (neu) with an aminoterminal signal sequence, a Pseudomonas exotoxin A translocation domain, and a constitutively active caspase-6 moiety. The strategy of using this immunotoxin is have this protein secreted by transfected cells, which then binds to tumor cells overexpressing the neu oncogen. Uptake of the construct causes caspase-6 to cleave lamin A, which leads to nuclear damage and causes tumor cell death by apoptosis
