This Drosophila melanogaster gene (CG4345) has been identified by Chen et al (1996) as a cell death activator protein. It was named grim as it is located in the vicinity of another gene, reaper.
The Grim protein contains two pro-apoptotic domains that are relevant for its killing function. The N-terminal region contains an RHG motif, which is required for disrupting blockage of caspase activity through interaction with IAP proteins. An internal GH3 domain, which triggers a mitochondrial pathway, is essential for Grim pro-apoptotic activity in mouse cells (Claveria et al, 2004).
