[ripoptosomes]
The ripoptosome constitutes a large protein complex (∼2 MDa) that is assembled in the cytosol and mediates both cell death by apoptosis and necroptosis in response to genotoxic stress and stimulation of TLR3 (Feoktistova et al, 2011; Tenev et al, 2011). This complex is necessary but not sufficient for cell death. The ripoptosome complex forms independently of TNF-alpha, CD95L (FAS ligand), TRAIL, death receptors, and mitochondrial pathways. It also forms upon SMAC mimetic treatment without involvement of autocrine TNF-alpha.
