Escherichia coli colibactin is a secreted hybrid non-ribosomal peptide-polyketide compound that induces DNA double-strand breaks in eukaryotic cells that then leads to cell cycle arrest, the progressive enlargement of cell bodies, and cell death (Nougayrède et al, 2006). A 54 kb genomic island, termed pks island, has been shown to be responsible for the synthesis of colibactin. This machinery consists of nonribosomal peptide megasynthases (NRPS), polyketide megasynthases (PKS), hybrid NRPS/PKS enzymes, different tailoring or editing enzymes, and a critical phosphopantetheinyl transferase that mediates NRPS and PKS activation.
