This domain of 50 amino acids is found in several Aminoacyl-tRNA synthetases. The name is derived from its occurrence in Tryptophanyl-tRNA synthetase (tryptophan = W), Histidyl-tRNA synthetase (histidine = W) and glutamyl-prolyl tRNA synthetase (EPRS).

Glycyl-tRNA synthetase, Histidyl-tRNA synthetase, methionyl-tRNA synthetase, and Tryptophanyl-tRNA synthetase contain a single WHEP domain.
