often also simply Vaccinia virus A46 protein or A46 protein or A46R protein or Poxvirus A46 protein. This protein, which contains a TIR domain, is encoded in the genome of Vaccinia virus and is found also in variola virus, the causative agent of smallpox (Stack et al (2005). A46R is expressed early during infection and contributes to Vaccinia virus virulence. Viruses lacking the A46R gene are attenuated in a murine intranasal model (Stack et al, 2005). The protein is a member of the Vaccinia virus BCL2 family and adopts a BCL2-like fold, similar to that seen with Vaccinia virus A52R protein but has evolved to inhibit NF-kappa-B rather than apoptosis
