Immunocasp-3

Immunocasp-3 is a chimeric tumor-targeted killer protein designed by Jia et al (2003). Immunocasp-3 comprises a single-chain antibody directed against erbB2 (neu) with an aminoterminal signal sequence, a Pseudomonas exotoxin A translocation domain, and a constitutively active caspase-3 moiety. The strategy of using this immunotoxin is to have this protein secreted by transfected cells, which then binds to tumor cells overexpressing the neu oncogen. Uptake of the construct and subsequent release of the caspase-3 moiety would lead to cell death by apoptosis.
