Hematopoietin receptor superfamily

Members of this family have conserved amino acid sequence motifs in the extracellular domain (4 positionally conserved cysteine residues and a conserved WSXWS. The cytoplasmic domains vary but in the membrane proximal domain one can observe conserved elements: Homology box 1 (proline rich motif) and Homology box 2 (less well defined, present in some family members not others).

Receptors consist of a cytokine-specific subunit (alpha chain) and a signal-transducing subunit (beta chain) required for high affinity binding of the cytokine. This signaling subunit is usually not specific for the cytokine.
