[Factor associated with N-SMase activation] Also: N-SMase activation-associated factor. The approved gene symbol for this factor is NSMAF [neutral sphingomyelinase activation-associated factor].
This ubiquitously expressed protein of 917 amino acids has been identified by Adam-Klages et al (1996). FAN directly interacts with the neutral sphingomyelinase (N-SMase)-activating domain (NSD) of the TNF-alpha receptor TNFR1 and couples the receptor to the N-SMase signaling pathway.
Segui et al (2001) have reported that stable expression in human fibroblasts of a dominant negative form of FAN abrogates TNF-induced ceramide generation from sphingomyelin hydrolysis and reduces caspase
