[caspase activation and recruitment domain; Caspase recruitment domain] A conserved sequence domain consisting of 6 or 7 antiparallel alpha helices found in a variety of adaptor proteins associated with cell surface receptors that signal cell death by apoptosis.
The CARD domain is engaged in homophilic interactions with the prodomains of cysteine proteases (caspases) and serves to recruit the inactive polypeptide precursors (zymogen) of these enzymes to the receptor-adaptor protein complex Hofmann et al, 1997).
CARD proteins have been shown to activate transcription factor NF-kappa-B and to interact with BCL10.
