[viral interferon regulatory factor-4] vIRF4 is encoded by an open reading frame in the genome of KSHV (KSHV orfK10) and has been detected by gene array analysis (Jenner et al, 2001). Unlike vIRF1, vIRF2, and vIRF3, vIRF4 is not engaged in modulating interferon responses. vIRF4 has been shown to deregulate the p53 tumor suppressor pathway by targeting MDM2 [Double minute 2 protein], a protein that has the function of an E3 ubiquitin-protein ligase and acts as a major regulator of the tumor suppressor p53 by targeting its destruction (Lee et al, 2009). Thus, vIRF4 may be involved in oncogenesis associated with KSHV infections.
