Semaphorins (approximately 780 amino acids) constitute a large and growing family of both secreted and transmembrane proteins. All semaphorins are characterized by an extracellular semaphorin domain of approximately 500 amino acids that has been conserved through metazoan evolution. There are 16 conserved cysteine residues. Secreted semaphorins have an immunoglobulin (Ig) domain C-terminal to the sema domain. Transmembrane semaphorins can have either an Ig domain or a thrombospondin type I repeat. Some transmembrane semaphorins do not have any structural motif C-terminal to their sema domain (Kolodkin et al, 1993).
