This term has been proposed for the process that preferentially removes damaged mitochondria from the cell before leading to cell death (Lemasters, 2005). As such, mitophagy is a specialized form of autophagy (mitochondrial autophagy).
The process is independent of cellular ATP collapse (Priault et al, 2005). Studies with yeast cells have shown that mitophagy is not a random but a selective process that requires a specific outer membrane protein, UTH1 (Kissova et al, 2004). Initiation of mitophagy may involve mitochondrial permeability transition (Rodriguez-Enriquez, 2006), but the exact molecular details of this process remain to be elucidated. The tightly regulated molecular mechanisms between mitochondrial biogenesis and mitophagy that ensure mitochondrial homeostasis have been reviewed by Palikaras and Tavernarakis, 2014.
