Tachycitin (73 amino acids) has been purified from hemocyte extracts of the horseshoe crab (Tachypleus tridentatus). Tachycitin inhibits the growth of both Gram-negative bacteria and Gram-positive bacteria, and fungi. Tachycitin and big Defensin act synergistically in antimicrobial activities. The aminoterminal 28 residues of tachycitin show sequence homology to a part of chitin-binding regions found in antifungal chitin-binding peptides, chitin-binding lectins, and chitinases, all of which have been isolated from plants. Tachycitin shows a specific binding to chitin but does not bind with the polysaccharides cellulose, mannan, xylan, and laminarin.
