[transmembrane protein 214] This human multipass transmembrane protein resides in the endoplasmic reticulum. Li et al (2013) have identified TMEM214 as a critical mediator of apoptosis caused by endoplasmic reticulum stress due to excessive aggregation of misfolded proteins, which causes the unfolded protein response and cell death if stress persists.
Overexpression of TMEM214 induces cell death by apoptosis, whereas knock-down of its expression has the opposite effect. TMEM214 is constitutively associated with the proform of caspase-4. TMEM214-induced apoptosis is abolished by a dominant negative mutant of procaspase-4, whereas apoptosis induced by caspase-4 is inhibited by
