RGD (Arginine-glycine-aspartate) was identified originally as a sequence motif within fibronectin responsible for mediating cell attachment. A variety of different proteins, including proteins of the extracellular matrix, contain this tripeptide sequence motif in extended, solvent accessible, and conformationally flexible loops. RGD functions as a cell adhesion and aggregation domain that is recognized by subsets of a family of cell surface receptors known as integrins (D'Souza et al, 1991; Ruoslahti, 1996), which thus link extracellular protein-protein interactions with the intracellular cytoskeleton of cells.
