An acronym of the amino acid sequence proline (P), glutamic acid (E), serine (S), and threonine (T). One or more regions rich in these four amino acids, which are often flanked by positively charged amino acids, are found in many eukaryotic proteins that undergo rapid degradation. It is thought that such regions serve as a signal for proteolytic degradation (Rechsteiner and Rogers, 1996; Rogers et al, 1986). PEST sequences in plasma membrane proteins often increase protein turnover by enhancing the binding of ubiquitin ligase, leading to ubiquitination, endocytosis, and proteasomal degradation (Roth et al, 1998).
