Vivier et al (1993) have identified PEN5 as an epitope expressed selectively on functionally differentiated CD56(dim) NK-cells. Cells expressing the antigen proliferate poorly in response to IL2, but are potent cytolytic effectors. Cells that do not express the antigen proliferate in response to IL2, but are poor cytolytic effectors. The antigen has been identified as an unusual sulfated poly-N-lactosamine carbohydrate related to keratan sulfate glycosaminoglycans.
PEN5 is expressed also in a developmentally regulated fashion in human and rat oligodendrocyte precursor cells and marks the transition from proliferative to postmitotic stages. In human
