[ovarian tumour-like protease domain] OTU is an acronym from ovarian tumor. In Drosophila melanogaster, ovarian tumors with hundreds of poorly differentiated germ cells are caused, among other things, by mutations in the otu gene (see for overview: King and Storto, 1988).
The Drosophila gene is the founder member of a superfamily of over 80 proteins predicted to be cysteine proteases. They contain conserved cysteine and histidine residues in a putative catalytic site (Makarova et al, 2000). OTU domains share a cysteine protease catalytic triad common to many deubiquitinating enzymes
