This protein is encoded in the genome of orf virus and has been shown to inhibit functions of transcription factor NF-kappa-B. Deletion of ORFV121 from the viral genome markedly decreased virulence and disease pathogenesis in sheep, indicating that ORFV121 is a virulence determinant in the natural host. ORFV121 has been shown to inhibit the phosphorylation and nuclear translocation of the NF-kappa-B p65 subunit. ORFV121 deletion mutants increased NF-kappa-B-mediated gene transcription, and the expression of ORFV121 in cell cultures significantly suppresses NF-kappa-B-regulated reporter gene expression.
