The gene encoding the murine protein has been cloned by Umemoto et al (2006). Four isoforms have been described, all of which contain a central mucin-like domain. Nepmucin is expressed on the luminal surface of high endothelial venules in lymph nodes, but not on high endothelial venules of Peyer patches. Umemoto et al (2006) have reported that the mucin-like domain of mouse nepmucin mediates lymphocyte rolling under physiologic conditions, which is dependent on L-selectin. Under static conditions, nepmucin binds to lymphocytes through its Ig domain, and promotes shear-resistant lymphocyte binding in combination with ICAM-1 under flow conditions.
