Metzincins

[Metzincin]

This name has been proposed for a superfamily of topologically similar proteolytic enzymes (zinc endopeptidases). Metzincins are mostly multidomain proteins with approximately 130-260-residue globular catalytic domains showing a common core architecture characterized by a long zinc-binding consensus motif, HEXXHXXGXX(H/D), and a methionine-containing Met-turn. These enzymes are, therefore, a subfamily members of a larger group of zinc-dependent enzymes, named zincins, characterised by a shorter consensus sequence, HEXXH, with the two histidines acting as ligands of the catalytic zinc and the glutamate as the general base.
