The synthetic tetrapeptide LEVD (Leu-Glu-Ala-Asp) conjugated to 7-amino-4-trifluoromethyl coumarin (LEVD-AFC) is a fluorogenic substrate for caspase-4 which is detectable following cleavage of the substituted coumarin moiety from the peptide. This synthetic inhibitor is available coupled to fluoromethylketone (LEVD-fmk) or the N-benzyloxycarbonyl-derivative (zLEVD-fmk).
Leu-Glu-Ala-Asp-aldehyde (LEVD-CHO) is a synthetic caspase-4 inhibitor.
