The synthetic peptide LEED (Leu-Glu-Glu-Asp) coupled to fluoromethylketone (zLEED-fmk) is a synthetic permeable inhibitor of caspase-13. LEED corresponds to residues 286-289 of caspase-13 which is the preferred substrate for caspase-8 cleavage, and caspase-8 is required to activate caspase-13 (Humke et al, 1998).
General note on chemical modifications: Practically all of the available caspase inhibitors, be they specific or pan-caspase inhibitors, come in chemically modified form. The introduction of an aldehyde group at the C-terminus of the short peptide sequences recognized by caspases
