Savino et al (1994) have generated a set of IL6 variants (see also: Muteins) that do no longer interact with the signal transducer gp130 and which that behave as potent cytokine receptor super-antagonists. These variants bind to the ligand-specific IL6 receptor-alpha chain with high affinity and inhibit wild-type cytokine activity (Sporeno et al, 1996). The most potent of these antagonists, Sant-7 is effective a variety of factor-dependent cell lines that require IL6.
In IL6 dependent XG-1 cells Sant-7 has been shown to cause cell death by apoptosis (Demartis et al, 1996).
