This protein is encoded by the open reading frame 4 in the E4 region of the adenovirus genome. The protein cooperates with the viral E1A protein and promotes cell death by apoptosis in the absence of p53 (Marcellus et al, 1996). Induction of apoptosis by adenovirus E4orf4 protein is specific to transformed cells (Shtrichman and Kleinberger, 1998) and requires an interaction with protein phosphatase 2A (Shtrichman et al, 1999). The protein activates caspase-8 and the FADD pathway in a cell line-specific manner (Livne et al, 2001).
