This enzyme, which catalyzes proteolytic degradation of PARP1 [poly(ADP-ribose) polymerase 1] in mammalian cells, has been purified by Nicholson et al (1995). The PARP cleavage protease consists of two subunits derived from a common proenzyme, which the authors have been identified as CPP32 [cysteine protease 32]. Fernandes-Alnemri et al (1996) have shown that the two subunits of Apopain are released from the proenzyme by granzyme B or the cysteine protease MCH4 (Caspase-10).
Nicholson et al (1995) have demonstrated also that an apopain peptide aldehyde inhibitor they developed prevents cell death by apoptosis
