This targeting peptide has been isolated in a phage display screen (Arap et al, 2004). The peptide binds to glucose-regulated protein-78 (GRP78), a clinically relevant tumor antigen expressed on the cell surface of metastatic tumors. Larson et al (2013) have used conjugates of this peptide and heat shock protein (HSP) targeted N-(2-hydroxypropyl)methacrylamide (HPMA) copolymer-drug conjugates in combination with aminohexylgeldanamycin (AHGDM) or docetaxel. HSP targeted docetaxel conjugates strongly inhibit human prostate cancer DU145 cells. HSP targeted AHGDM and docetaxel conjugates show synergistic effects in combination with hyperthermia.
