[tripeptidylpeptidase 2; tripeptidylaminopeptidase 2] This enzyme has been isolated by Balow et al (1983). It removes tripeptides from the N-terminus of longer peptides at neutral pH. The enzyme copurifies with 26S proteasomes, may degrade proteins in conjunction with proteasomes but also act as a nonproteasomal enzyme (Geier et al, 1999).
Stavropoulou et al (2006) have reported that overexpression of the enzyme correlates with accelerated growth and the appearance of centrosome and chromosome aberrations, suggesting an involvement in the induction and/or maintenance of genetic instability in malignant cells. TPP2 appears to promote malignant cell growth by allowing cells to exit from mitosis, survive with severe mitotic spindle damage, and escape from cell death by apoptosis.
