This enzyme (EC2.4.2.12), known also as NMN diphosphorylase or NMN pyrophosphorylase (abbr. NAmPRTase, Nampt) catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, an intermediate in the biosynthesis of NAD. This is the rate limiting component in the mammalian NAD biosynthesis pathway. The enzyme can regulate cellular levels of NAD and so impact not only cellular energetics but also NAD-dependent enzymes.
The abbr. iNampt and eNampt are used to denote intracellular and extracellular forms of the enzyme, respectively. The extracellular form of the enzyme has been identified independently as a cytokine, PBEF
