These highly TNF-sensitive tumorigenic murine fibroblasts are used to assay TNF-alpha and TNF-beta (see also: bioassays, cytokine assays) by measuring the degree of cell lysis induced by these factors in the presence of actinomycin D under serum-free conditions. Both factors can be distinguished from each other by using appropriate neutralizing antibodies. Recombinant TNF-beta (see also: Recombinant cytokines) expressed in Escherichia coli is also cytolytic for L-M cells. Oligomeric TNF is cytotoxic towards L-M cells, whereas monomeric TNF is virtually inactive. L-M cells are 3 to 4 times more sensitive to cytolysis by
