H16 peptide (Iwasaki et al) is a polyhistidine peptides of 16 histidine residues (HHHHHHHHHHHHHHHH). H16 peptide is a cell-penetrating peptide that shows higher cell-penetrating capacity against HT1080 human fibrosarcoma cells octa-arginine peptide. Cellular uptake of H16 peptide is due mainly to macropinocytosis and most of the H16 peptide localizes in the lysosome and Golgi apparatus. A cytoplasmic pro-apoptotic domain (KLAKLAKKLAKLAK) conjugated to the H16 peptide shows cytotoxic effects. In vivo distribution analysis shows that the H16 peptide is a tumor targeting peptide that accumulates immediately in tumor tissue and is retained up to 132h following injection into the tumor (HT1080
