[glutathione peroxidase 4] GPX4 is a ubiquitously expressed (Baek et al, 2007) selenoprotein oxidoreductase enzyme (197 amino acids, 19 kDa) that associates with membranes, cytoplasm, and mitochondria (Januel et al, 2006) and directly reduces phospholipid hydroperoxides within membranes and lipoproteins, using glutathione as a cofactor. It acts in conjunction with alpha-tocopherol (vitamin E) to inhibit lipid peroxidation (Scheerer et al, 2007). Only GPX4 reduces cholesterol and complex membrane lipid hydroperoxides (Ursini et al, 1985; Thomas et al, 1990).
The human cDNA has been cloned by Esworthy et al (1994), who referred to it as PHGPX [Phospholipid hydroperoxide glutathione peroxidase]. Kelner and Montoya (1998) have reported the structure of the human GPX4 gene.
