DRAM has been identified as a p53 target gene encoding a lysosomal protein that induces cell death by macroautophagy by acting as an effector of p53-mediated cell death (Crighton et al, 2006). Overexpression of DRAM alone causes minimal cell death, but DRAM is essential for p53-mediated apoptosis. The analysis of primary tumors has shown that expression of DRAM is frequently decreased and that this is often accompanied by retention of wild-type p53.
