Internucleosomal DNA fragmentation is one of the hallmarks of cells dying from apoptosis. Under suitable conditions this produces a ladder of DNA fragments during gel electrophoresis (Yeung et al, 2002). The assay depends upon the degree of apoptosis and the number of cells. If DNA laddering can be detected this supports morphological or other criteria usually taken as evidence for the occurrence of apoptotic cell death.
It should be noted that an orderly pattern of DNA fragmentation is observed also as a consequence of freezing even when caspases- the critical enzymes involved in apoptotic cell death, are blocked (Kok et al, 2002).
