Megakaryocytes are the largest normally occuring cells in the marrow. They arise from pluripotent stem cells, undergo endomitosis (DNA duplication without cell division) and thus become polyploid during amplification (usually 16N with a range in ploidy of 4N - 64N). The process of their formation is called megakaryocytopoietis. In the presence of thrombopoietin, megakaryocytes mature by polyploidization and cytoplasmic maturation, undergo drastic morphological changes, and form proplatelet, which release a number of mature platelets.
A study of the global expression profile of human megakaryocytes has been reported by Shim et al (2004).
