The synthetic peptide VDVAD (Val-Asp-Val-Ala-Asp) conjugated to 7-amino-4-trifluoromethyl coumarin (VDVAD-AFC) is a fluorogenic substrate for caspase-2 which is detectable following cleavage of the substituted coumarin moiety from the peptide (Talanian et al, 1997). Although caspase-2 efficiently cleaves peptides containing the sequence VDVAD, caspase-3 can cleave these peptides with similar kinetics (Talanian et al, 1997).
