The U-box is a modified RING finger motif found in various eukaryotic proteins (Johnson et al, 1995; Koegl et al, 1999; Aravind and Koonin, 2000). The U-box is a domain of approximately 70 amino acids without the full complement of Zn ion binding ligands. The U-box domain can interact directly with several E2 enzymes of the ubiquitination pathway and is thought to function similar to the RING finger motif in the ubiquitin-conjugating system (Pringa et al, 2001). The U-box is of ancient origin (Marin, 2010), and members of the U-box family of proteins constitute a distinct class of ubiquitin-protein ligases.
