[stromal cell-derived inducing activity] This biochemically uncharacterized activity is found to accumulate on the cell surface of the stromal cell line PA6 (Kawasaki et al, 2000). Coculture of murine embryonic stem cells with PA6 cells induces neuronal differentiation of ES cells in serum-free medium without use of either retinoic acid or embryoid bodies. Neuralization is suppressed by BMP4, which promotes epithelial differentiation. This coculture system yields a high proportion of neurons expressing tyrosine hydroxylase and producing dopamine.
