LEHD

The synthetic tetrapeptide LEHD (Leu-Glu-His-Asp) conjugated to 7-amino-4-trifluoromethyl coumarin (LEHD-AFC) or 7-amino-4-methyl coumarin (LEHD-AMC) are fluorogenic substrates for caspase-1 and caspase-9 which are detectable following cleavage of the substituted coumarin moiety from the peptide. LEHD conjugated to p-nitroanilide (LEHD-pNA) is a colorimetric substrate for caspase-9 enzyme activity. zLEHD-fmk is the N-benzyloxycarbonyl-LEHD-methyl-fluoromethylketone derivative. An enzyme that recognizes the LEHD sequence is referred to sometimes also as LEHDase.
