The ITSM motif is a stretch of amino acids (TxYxxV/I) identified by virtue of its ability to bind to the small adaptor SH2D1A [Src homology 2 domain-containing molecule 1A] (SAP; Signaling lymphocytic activation molecule-associated protein, which is defective in X-linked lymphoproliferative disease) (Sidorenko and Clark, 2003). The ITSM, defined originally in CD150, recruits the tyrosine phosphatase SHP-2 [Src homology region 2 domain-containing phosphatase-2] to the CD150 cytoplasmic tail.
