This element has been described originally as a sequence in the promoter region of TNF-alpha that mediates mRNA decay independent of ARE [AU-rich element], another sequence mediating mRNA decay (Stoecklin et al, 2003). The conserved constitutive decay element represses TNF-alpha expression in macrophages by maintaining the mRNA short-lived, thereby preventing excessive induction of TNF-alpha after LPS stimulation. Subsequent work has shown that many other mRNAs encoding regulators of development and inflammation contain this element (Leppek et al, 2013) and are subject to degradation in a process that involves the ROQ domain of RNA binding proteins such as
