[alpha-1-purothionin] alpha-1-PTH (Jones and Mak, 1977) has been isolated from wheat endosperm of Triticum aestivum sp. and has been suggested to play a role in plant defence mechanisms against phytopathogenic organisms. The cDNA has been cloned by Castagnaro et al (1994).
alpha-1-PTH forms channels with the following selectivity Ca(2+) > Na(+) approximately K(+) >> Cl(-) and Cl(-) >> Na(+). In isolated mouse neuromuscular preparations, alpha-1-PTH induces muscle membrane depolarization, leading to blockade of synaptic transmission and directly elicits muscle twitches. alpha-1-PTH also causes swelling of differentiated neuroblastoma NG108-15 cells, membrane bleb formation, and disorganization of F-actin. (Lianos et al, 2006).
