The commercially available aldefluor assay is based on the conversion of the ALDH substrate, BODIPY-aminoacetaldehyde (4,4 difluoro-4-bora-3a,4a-diaza-s-indacene-aminoacetaldehyde) to the fluorescent reaction product BODIPY-aminoacetate. This reaction is catalyzed by aldehyde dehydrogenases.
It is generally assumed that the predominant enzyme is ALDH1, but Moreb et al (2012) have reported that the assay is not specific for ALDH1. Nevertheless, the detection of high level ALDH1 dehydrogenase activity in living cells using this assay correlates with cells possessing the properties of stem cells or cancer stem cells, and monitoring this activity can be used to identify and enrich such cells (see:
