[aggrecanase]
a collective term for proteolytic enzymes that cleave the core protein of the extracellular matrix molecule aggrecan. Several members of the ADAMTS protein family have been shown to cleave the Glu373-Ala374 bond of the aggrecan protein.
Metalloproteinases with aggrecanase activity are ADAMTS1, ADAMTS4, ADAMTS5, ADAMTS8, ADAMTS9, ADAMTS15.
ADAMTS4 and ADAMTS5 in particular play a major role in early stages of cartilage destruction in rheumatoid arthritis and osteoarthritis, and their inhibition prevents aggrecan degradation in diseased cartilage (Nagase and Kashiwagi, 2003; Le Maitre et al, 2007).
