ORF61, encoded by an open reading frame in the genome of Varicella zoster virus is an immediate early gene expressed during infection with Varicella zoster virus. The protein down-regulates the stimulation of viral putative immediate-early, early, and late promoters by other viral trans-activator genes (Nagpal S and Ostrove, 1991). ORF61 can trans-activate or trans-repress the transcription of other viral proteins, including its own promoter, to enhance infectivity (Moriuchi et al, 1993, 1994; Wang et al, 2009; Perera et al, 1992). Zhu et al (2011) have reported that overexpression of ORF61 has only a minor effect on Sendai virus-induced NF-kappa-B signalling. ORF61 interacts with phosphorylated IRF3 and induces degradation of the protein, thereby preventing IFN-beta expression. The ORF61 protein contains an N-terminal RING domain
