[TUBBY domains]
The Tubby domain comprises approximately 260 amino acids and was identified first in the tubby protein implicated in mature-onset obesity (Santagata et al, 2001). The domain is capable of interacting with DNA and phosphotidylinositol and therefore is functioning as a membrane-targeting domain. The Tubby domain of the tubby and TULP proteins (proteins that contain a TUBBY domain) (Carroll et al, 2004; Mukhopadhyay and Jackson, 2011) binds with high specificity to biphosphorylated phosphoinositides phosphorylated at the 4-position on the inositol ring. Hydrolysis of PI(4,5)P2 by PLC releases the domain from the membrane. It tranlocates into the nucleus, binds DNA, and allows the tubby protein amino-terminal transcription factor-like activation domain
