Genetically engineered variants of TNF-alpha (see also: Muteins). A derivative of TNF-alpha, designated as F4236, has been described by Miyata et al (1992). This protein carries a cell-adhesive sequence (Tyr-Ile-Gly-Ser-Arg) at the N-terminus. Unlike the wild-type compount F4236 exhibit antitumor activity in fibrosarcoma- and melanoma-bearing mice but does not enhance metastasis.
Shikama et al (1995) have described another derivative, F4614, also carrying a cell-adhesive sequence and another point mutation. The protein displays considerably reduced hypotensive effects. It also shows a higher efficacy in MethA fibrosarcoma-transplanted mice than wild-type TNF-alpha.
