[ROC domains]
[Ras of complex proteins domain] This domain has been observed in a group of multidomain proteins, mostly kinases, known as ROCO proteins, which are found in both prokaryotes and eukaryotes (Marín et al, 2008; Klein et al, 2009). The ROC is a GTPase domain similar to Ras and other small G-proteins. In the ROCO proteins it is followed by a COR domain [C-terminal of ROC domain]. GTP binding to the ROC domain activates kinase activity. In human LRRK2 [leucine-rich repeat kinase 2], mutations that interfere with GTP hydrolysis lead to a hyperactivated kinase. Mutations within the COR domain of LRRK2 affect GTPase activity (Marín et al, 2008; Daniels et al, 2011).
